Editors Choice Article | Mitotherapy as a Novel Therapeutic Strategy for Mitochondrial Diseases

Background: The mitochondrion is a multi-functional organelle that is mainly responsible for energy supply in the mammalian cells. Over 100 human diseases are attributed to mitochondrial dysfunction. Mitochondrial therapy (mitotherapy) aims to transfer functional exogenous mitochondria into mitochondria-defective cells for recovery of the cell viability and consequently, prevention of the disease progress.


Objective: The review summarizes the evidence on exogenous mitochondria that can directly enter mammalian cells for disease therapy following local and intravenous administration, and suggests that when healthy cells donate their mitochondria to damaged cells, the mitochondrial transfer between cells serve as a new mode of cell rescue. Then the transferred mitochondria play their roles in recipient cells, including energy production and maintenance of cell function.
Conclusion: Mitotherapy makes the of modulation of cell survival possible, and it would be a potential therapeutic strategy for mitochondrial diseases.

Background: This study aimed to verify the influence of different substrates with and without addition of nutrient solution on the roostocks production of Capdeboscq cultivar and grafted peach plants of ‘Chimarrita’ scion.

Methods: In the first experiment, the height and stem diameter of the rootstocks were evaluated every two weeks, up to 90 days after transplanting (DAT). At 90 DAT, dry weight of shoots and roots, total dry mass and Dickson Quality Index were evaluated. In the second experiment, seedlings of ‘Capdeboscq’ were grafted with ‘Chimarrita’ scion. The growth of the scions and the percentage of living grafts were evaluated. At 146 DAT, the stem diameter of the scions, the SPAD index, the chlorophyll and nitrogen balance index were evaluated.

Discussion: The greatest mean values for the stem diameter of seedlings of the cv. Capdeboscq were obtained with the substrates T4 (5.53 mm); T2 (5.47 mm) and T1 (5.23 mm) with addition of nutrient solution, with seedlings reaching the plant standards according to the ordinance number 173 of May 27th of 1984, which recommends that rootstocks have to have a minimum stem diameter of 5.0 mm. Thus, the substrates which received the addition of nutrient solution, except the soil substrate, were adequate considering the rules that governing the peach tree production in Brazilian Nurserioes.

Conclusion: The nutrient solution already avaliblabe for Recent patents is highly indicated to obtain seedlings of ‘Capbdboscq’ rootstocks with high mophophysiological quality in less time during nursery cycle of plant production. The largest stem diameter for ‘Capdeboscq’ was obtained with substrate 35% sand + 15% soil + 50% bovine manure (5.53 mm); 75% sand + 25% soil (5.47 mm) and 100% sand (5.23 mm) with addition of nutrient solution. The best morphophysiological characteristics of ‘Chimarrita’ plants grafted on ‘Capdeboscq’ seedlings was obtained with the use of substrates 75% sand + 25% soil and 35% sand + 15% soil + 50% bovine manure, whose plants reached a morphological standard for sale, in just four and a half months DAT, that is 56 days after grafting.

Background: The success story of liposomes in the treatment of systemic infectious diseases and various carcinomas lead the scientists to the innovation of elastic vesicles to achieve similar success through transdermal route. In this direction, ethosomes and transfersomes were developed with the objective to design the vesicles that could pass through the skin. However, there is a lack of systematic review outlining the principles, method of preparation, latest advancement and applications of ethosomes and transfersomes. This review covers various aspects that would be helpful to scientists in understanding advantages of these vesicular systems and designing a unique nano vesicular delivery system.

Methods: Structured search of bibliographic databases for previously published peer-reviewed research papers was explored and data was culminated in terms of principle of these vesicular delivery systems, composition, mechanism of actions, preparation techniques, methods for their characterization and their application.

Results: A total of 182 papers including both, research and review articles, were included in this review in order to make the article comprehensive and readily understandable. The mechanism of action and composition of ethosomes and transfersomes was extensively discussed. Various methods of preparation such as, rotary film evaporation method, reverse phase evaporation method, vortex/ sonication method, ethanol injection method, freeze thaw methods, along with their advantages has been discussed. It was also discussed that both these elastic nanocarriers offer unique advantages of ferrying the drug across membranes, sustaining drug release as well as protecting the encapsulated bio actives from external environment. The enhanced bioavailability and skin penetration of ethosomes as compared to conventional vesicular delivery systems is attributed to the presence of ethanol in the bilayers while that for transfersomes accrues due to their elasticity along with their ability to retain their shape because of the presence of edge activators. Successful delivery of synthetic drugs as well as phytomedicines has been extensively reported through these vesicles.
Conclusion: Though these vesicular systems offer a good potential for rational drug delivery, a thoughtfully designed process is required to optimize the process variables involved. Industrial scale production of efficacious, safe, cost effective and stable formulations of both these delivery systems appears to be a pre-requisite to ensure their utility as the trans-dermal vehicles.
