Abstract

Introduction: Hypertriglyceridemia (HTG) is one of the most common metabolic disorders leading to pancreatitis and cardiovascular disease. HTG develops mostly due to impaired metabolism of triglyceride-rich lipoproteins. Although monogenic types of HTG exist, most reported cases are polygenic in nature.

Aim: This review article is focused on the classification of Primary HTG and the genetic factors behind its development with the aim of providing clinicians a useful tool for early detection of the disease in order to administer proper and effective treatment.

Discussion: HTG is often characterized by a complex phenotype resulting from interactions between genetic and environmental factors. In many instances, the complexity, perplexing causes, and classification of HTG make it difficult for clinicians to properly diagnose and manage the disorder. Better availability of information on its pathophysiology, genetic factors involved, environmental causes, and their interactions could help in understanding such complex disorders and could support its effective diagnosis and treatment.

Conclusion: The current review has summarized the case definition, epidemiology, pathophysiology, clinical presentation, classification, associated genetic factors, and scope of genetic screening in the diagnosis of primary HTG.

Abstract:

Background: With the rapid development of biological research, microRNAs (miRNAs) have increasingly attracted worldwide attention. The increasing biological studies and scientific experiments have proven that miRNAs are related to the occurrence and development of a large number of key biological processes which cause complex human diseases. Thus, identifying the association between miRNAs and disease is helpful to diagnose the diseases. Although some studies have found considerable associations between miRNAs and diseases, there are still a lot of associations that need to be identified. Experimental methods to uncover miRNA-disease associations are time-consuming and expensive. Therefore, effective computational methods are urgently needed to predict new associations.

Methodology: In this work, we propose an integrated method for predicting potential associations between miRNAs and diseases (IMPMD). The enhanced similarity for miRNAs is obtained by combination of functional similarity, gaussian similarity and Jaccard similarity. To diseases, it is obtained by combination of semantic similarity, gaussian similarity and Jaccard similarity. Then, we use these two enhanced similarities to construct the features and calculate cumulative score to choose robust features. Finally, the general linear regression is applied to assign weights for Support Vector Machine, K-Nearest Neighbor and Logistic Regression algorithms.

Results: IMPMD obtains AUC of 0.9386 in 10-fold cross-validation, which is better than most of the previous models. To further evaluate our model, we implement IMPMD on two types of case studies for lung cancer and breast cancer. 49 (Lung Cancer) and 50 (Breast Cancer) out of the top 50 related miRNAs are validated by experimental discoveries.

Conclusion: We built a software named IMPMD which can be freely downloaded from https:// github.com/Sunmile/IMPMD.

Objective and Conclusion: This review describes methods for the production of 225Ac and its daughter nuclide 213Bi and summarizes the current clinical experience with both alpha emitters with particular focus on recent studies of targeted alpha therapy of bladder cancer, brain tumors, neuroendocrine tumors and prostate cancer.

Abstract:

We present a method with fusion of images of three modalities 18F-FDG PET, CT, and 3-D ultrasound (US) applied to imaging of the anal canal and the rectum. To obtain comparable geometries in the three imaging modalities, a plexiglas rod, with the same dimensions as the US transducer, is placed in the anal canal prior to the PET-CT examination. The method is based on manual co-registration of PET-CT images and 3-D US images. The three-modality imaging of the rectum-anal canal may become useful as a supplement to conventional imaging in the external radiation therapy in the treatment of anal cancer, where the precise delineation of a tumor is crucial to avoid damage from radiation therapy to the healthy tissue surrounding it. The technique is still in a phase of development, and the demands for integration different company software systems are significant before commercial application. Three-modality imaging may also be used in certain other diagnostic or therapeutic fields.

Stem Cell Drug May Halt Cancer Spread

Southern California scientists may have found a potential cancer breakthrough thanks to new stem cell research at the University of California, Los Angeles.

Stem cells — usually studied for their disease-curing potential by stimulating growth or replacing diseased cells — are being examined in a different way by researchers. This time, scientists are looking at how they can block bad stem cells that may lead to fatal cancers.

The cells are called “stem cells” because they are the most immature cells that are taken from a baby’s umbilical cord, from bone marrow, or even from an embryo.

Turning Stem Cells into a Powerful Weapon Against AIDS

At UCLA’s special stem cell laboratory in West Los Angeles, scientists have received a grant to see if different types of stem cells can be blocked so that a cancer may be controlled, or even cured.

“Within cancers, there are stem cells that promote the growth of additional cancer cells,” said Dr. Zeb Wainberg, of UCLA’s Jonsson Comprehensive Cancer Center (JCCC).

That discovery led to research on a unique, cancer-fighting drug that worked on the cancerous stem cells.

“The drug actually blocks the enzyme activity of an enzyme that’s critical in cell division,” said Dr. Dennis Slamon of UCLA’s JCCC.

In the lab and in tests on animals, the resulting tumors were contained and prevented from spreading.

Scientists believe the next step is to start trials in humans to see if the drugs work the same way as they did in the lab, and to see what the effect is on cancer in humans.

Stem Cells Help Reconstruct Cancer Patient’s Breast

Slamon and Wainberg may be able to start those trials as early as this April, thanks to a grant from the California Institute for Regenerative Medicine, the state stem cell agency.

“We will be testing that drug in humans for the first time,” Wainberg said. “This treatment will be used for patients that we believe have colorectal cancer, breast cancer, and possibly some forms of brain center.”

“The way you’ll know if the treatment has worked is you will see responses in the tumor – that is, shrinkage of the tumor – and control of growth over time so that you don’t see progression of the tumor,” Wainberg said.

The hope is that the trial will lead to a cure for some patients and control of cancer for others, with fewer side effects than conventional treatments.

The study will also help determine which cancers and which patients the treatment would best suit.
